Drug intelligence / Profile preview
This is a combination therapy of **zandelisib**, **venetoclax**, and **rituximab** under investigation for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL)[1]. - **Zandelisib** is a selective phosphatidylinositol 3-kinase delta (PI3Kδ) inhibitor, designed to block B-cell receptor signaling and decrease malignant B-cell proliferation and survival. - **Venetoclax** is a small molecule BCL-2 inhibitor that induces apoptosis in CLL cells by mimicking BH3-only proteins, thus promoting cell death in malignant lymphocytes[2][4][5]. - **Rituximab** is a monoclonal antibody targeting the CD20 antigen on B lymphocytes, leading to cell lysis through complement- and antibody-dependent mechanisms[2][4][5]. This regimen is being studied in phase 2 clinical trials for adult patients with relapsed or refractory CLL, especially those who have received at least one prior line of therapy[1]. Clinical rationale for the combination is based on complementary mechanisms of action disrupting CLL cell survival and proliferation.
