**Zemprocitinib (LNK01001)** is a highly selective, oral small molecule Janus kinase 1 (JAK1) inhibitor developed primarily by Lynk Pharmaceuticals, with involvement from Simcere. It exhibits potent JAK1 inhibition (IC50 5.95 nM) with >20-fold selectivity over JAK2 (141.3 nM) and TYK2 (119 nM), targeting autoimmune and inflammatory diseases by blocking JAK1-mediated signaling pathways involved in cytokine-driven inflammation. Positive Phase 2 data support efficacy in moderate-to-severe atopic dermatitis (significant EASI improvements), rheumatoid arthritis (ACR20/50 responses), and ankylosing spondylitis (ASAS40 endpoint), with good tolerability and mild-to-moderate adverse events; it is advancing in multiple Phase 3 trials in China.
