ZI-KL-1 is an allogeneic, off-the-shelf T cell receptor-engineered natural killer (TCR-NK) cell therapy being developed by Zelluna Immunotherapy for the treatment of various solid tumors. The therapy consists of natural killer cells engineered with a T cell receptor (TCR) specific for the Kita-Kyushu lung cancer antigen 1 (KK-LC-1), a cancer-testis antigen that is widely expressed across several solid malignancies—including breast, gastric, lung, pancreatic, and cervical cancers—while maintaining very limited distribution in normal tissues. By combining the innate cytotoxic mechanisms of NK cells with the precise tumor-targeting capability of TCRs, ZI-KL-1 is designed to overcome current challenges in treating solid tumors, such as antigen escape and the immunosuppressive tumor microenvironment. The KK-LC-1 TCR was exclusively licensed from the U.S. National Cancer Institute (NCI) in 2021.
