Overview

ZTx102 is a novel, first-in-class small molecule inhibitor being developed by Zehna Therapeutics for the treatment of Chronic Kidney Disease (CKD). The drug targets the gut microbial enzyme CutC lyase (choline trimethylamine lyase), which converts dietary choline to trimethylamine (TMA), a precursor of trimethylamine N-oxide (TMAO). High systemic levels of TMAO have been linked to accelerated development of both cardiovascular and chronic kidney disease. ZTx102 is formulated as an orally administered, colon-directed formulation designed to minimize systemic absorption in the upper gastrointestinal tract and maximize delivery to the colon where CutC-containing bacteria are located. In preclinical studies using a rat adenine-induced model of CKD, ZTx102 demonstrated the ability to normalize elevated serum TMAO levels, slow the loss of kidney function, reduce serum creatinine levels, slow eGFR decline, and reduce biomarkers of kidney fibrosis and myocardial stress.
