Overview

The ZSDT regimen is a combination therapy consisting of zuberitamab, selinexor, dexamethasone, and thalidomide. It is currently being evaluated in clinical trials led by Zhengzhou University for the treatment of patients with relapsed or refractory mantle cell lymphoma (MCL) who have previously failed treatment with Bruton's tyrosine kinase (BTK) inhibitors. The regimen combines several distinct therapeutic modalities: zuberitamab is a monoclonal antibody targeting CD20; selinexor is a first-in-class selective inhibitor of nuclear export (SINE) that blocks Exportin 1 (XPO1); dexamethasone is a corticosteroid that acts as a glucocorticoid receptor agonist; and thalidomide is an immunomodulatory drug (IMiD) that targets the E3 ubiquitin ligase cereblon. This multi-targeted approach is designed to enhance anti-tumor efficacy through synergistic mechanisms while potentially reducing toxic side effects compared to traditional chemotherapy.
