Overview

ZYSM003 is a small molecule Pan-Ras(ON) inhibitor being developed by Rongchang Pharmaceuticals for the treatment of solid tumors. Unlike traditional KRAS G12C inhibitors that bind to the inactive OFF state (GDP-bound), Pan-Ras(ON) inhibitors are designed to target the active ON state (GTP-bound) of various Ras isoforms, including KRAS, NRAS, and HRAS. This approach potentially addresses a broader range of Ras mutations and overcomes resistance mechanisms associated with state-specific inhibition. The drug is currently in the IND-enabling stage of development.
