Overview

Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen on the surface of pre-B and mature B lymphocytes. By binding to CD20, rituximab induces B-cell lysis through mechanisms such as complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC), leading to depletion of both normal and malignant B cells while sparing plasma cells and hematopoietic stem cells. It is primarily used in the treatment of non-Hodgkin lymphoma, chronic lymphocytic leukemia, rheumatoid arthritis (in patients with inadequate response to TNF inhibitors), granulomatosis with polyangiitis, microscopic polyangiitis, and pemphigus vulgaris. Rituximab was originally developed by IDEC Pharmaceuticals in collaboration with Genentech[1][5][7].
