Overview

Antibody-coated cancer cells represent a biological state where tumor cells have been opsonized by therapeutic monoclonal antibodies. This coating serves as a molecular bridge between the malignant cell and the host's immune system, specifically through the interaction of the antibody's Fc region with Fc receptors on effector cells such as Natural Killer (NK) cells and macrophages (Nimmerjahn & Ravetch, 2008). This interaction facilitates various effector functions, including antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC) (Wang et al., 2015). While not a single molecular target itself, the formation of these complexes is the fundamental requirement for the efficacy of many immunotherapy agents. The success of this targeting strategy depends on the density of the surface antigen, the affinity of the antibody, and the presence of a functional immune microenvironment. Clinically, this mechanism is exploited by numerous approved drugs, such as rituximab for B-cell lymphomas and trastuzumab for HER2-positive breast cancer (Weiner, 2015). However, therapeutic challenges include the potential for off-target effects on healthy cells expressing the same antigen and the development of resistance through antigen shedding or downregulation.
