Epidermal Growth Factor Receptor 2 (HER2) Extracellular Domain 4 is a subdomain within the extracellular region of the HER2 receptor. It is involved in receptor dimerization and activation processes. HER2 overexpression, and in particular the accessibility and confirmation of the extracellular domains, is strongly implicated in various cancers, especially breast and gastric cancers, making this domain a target for therapeutic intervention. Monoclonal antibodies, such as trastuzumab, bind near domain IV, inhibiting dimerization/activation and flagging cells for immune-mediated destruction.
