Overview

ERBB2 (HER2) is a receptor tyrosine kinase and a member of the epidermal growth factor receptor (EGFR) family. It does not bind ligand directly but is activated through homo- or heterodimerization with other EGFR family members, leading to downstream signaling cascades that regulate cell proliferation, survival, differentiation, and migration. Overexpression or amplification of ERBB2 is associated with aggressive tumor behavior and poor prognosis in several cancers, particularly breast cancer, making it an important biomarker and therapeutic target.
