HLA-E is a non-classical MHC class I molecule that plays a crucial role in immune regulation. It presents peptides, primarily derived from the leader sequences of other MHC class I molecules, to CD94/NKG2 receptors on NK cells and some CD8+ T lymphocytes. Binding to these receptors can either inhibit or activate NK cell responses, thereby balancing innate immunity with adaptive memory responses. Tumors can exploit high HLA-E expression to evade immune surveillance by inhibiting NK cell-mediated lysis.
