Overview

Host immune and inflammatory pathways encompass the complex network of molecular and cellular interactions that mediate the body's response to injury and infection (National Institute of Allergy and Infectious Diseases, 2023). These pathways involve diverse components, including pattern recognition receptors (PRRs), cytokines, and intracellular signaling cascades like the NF-kB and JAK-STAT pathways (Nature Reviews Immunology, 2021). Dysregulation of these pathways is central to the pathogenesis of autoimmune diseases, chronic inflammatory conditions, and the 'cytokine storm' observed in severe infections (PubMed, 2020). Because this term describes a broad biological system rather than a single molecular entity, it is not considered a specific therapeutic target in the traditional sense. Instead, drug discovery efforts focus on modulating individual nodes within these pathways, such as specific interleukins or kinases, to achieve therapeutic effects while minimizing systemic toxicity (StatPearls, 2023).

Mechanism of action

Drugs modulate these pathways by targeting specific nodes such as cytokines, cell surface receptors, or intracellular signaling enzymes (e.g., kinases) to either suppress overactive inflammation or enhance deficient immune responses (StatPearls, 2023).
