Overview

The human epidermal growth factor receptor (HER/ErbB) family consists of four closely related transmembrane tyrosine kinase receptors: HER1 (EGFR/ErbB1), HER2 (ErbB2/neu), HER3 (ErbB3), and HER4 (ErbB4). Dimerization among these receptors—either as homodimers or heterodimers—is a critical event for their activation, leading to downstream signaling that regulates cell proliferation, survival, differentiation, and other processes relevant to cancer biology. Overexpression/amplification of these receptors—especially HER2—is implicated in various cancers and serves as a therapeutic target.
