Human Epidermal Growth Factor Receptor Type 2 (HER2) is a receptor tyrosine kinase and a member of the ErbB family. Unlike other ErbB receptors, HER2 has no known natural ligand and is primarily activated through dimerization, particularly heterodimerization with HER3. Activation of HER2 triggers downstream signaling pathways such as MAPK and PI3K/Akt, promoting cell proliferation and survival. HER2 overexpression or amplification is observed in various cancers, making it a significant prognostic/predictive marker and a therapeutic target. Anti-HER2 therapies have improved outcomes in HER2-positive cancers.
