HER4 (Human Epidermal Growth Factor Receptor 4) is a receptor tyrosine kinase and a member of the EGFR/HER/ErbB family. It plays a critical role in various physiological functions, including cardiac development, central nervous system development, mammary gland differentiation, and regulation of gene transcription. It is activated by ligands such as neuregulins, betacellulin, epiregulin and HB-EGF, leading to dimerization and activation of downstream signaling pathways like MAPK/ERK and PI3K/Akt. Its role in cancer is complex and context-dependent, with studies suggesting both tumor-suppressive and pro-proliferative functions. Mutations and SNPs have been associated with schizophrenia and ALS. Disruption of HER2/HER4 signaling is implicated in cardiotoxicity.
