Overview

Human innate immune and inflammatory pathways represent the fundamental biological processes responsible for the immediate detection and response to harmful stimuli, such as pathogens, damaged cells, or irritants (StatPearls, 2023). These pathways utilize various pattern recognition receptors, including Toll-like receptors and NOD-like receptors, to trigger intracellular signaling cascades like the NF-κB and inflammasome pathways (NIH, 2022). The resulting production of cytokines, chemokines, and antimicrobial peptides is essential for host survival but can lead to tissue damage if not strictly regulated. Dysregulation of these pathways is central to the pathogenesis of chronic inflammatory diseases, autoimmune disorders, and even certain cancers (Nature Reviews Immunology, 2017). Therapeutic agents often target specific components of these pathways, such as TNF-alpha or Janus kinases, to mitigate excessive inflammation. However, because these pathways are critical for defense, their inhibition can lead to significant safety concerns, most notably an increased risk of serious and opportunistic infections.
