Overview

The immune and inflammatory pathway network is a complex, integrated system of signaling cascades and cellular interactions that coordinate the host response to pathogens and tissue injury (Nature Reviews Immunology, 2021). This network includes major signaling hubs such as the NF-κB, JAK-STAT, and MAPK pathways, which regulate the production of cytokines, chemokines, and other inflammatory mediators (NIH, 2023). While essential for host defense, chronic or excessive activation of these pathways is a primary driver of autoimmune diseases, chronic inflammatory conditions, and cancer progression (StatPearls, 2023). Because it represents a broad biological system rather than a single molecular entity, it is not considered a specific therapeutic target in the traditional sense. Instead, drug discovery efforts focus on modulating specific nodes within the network, such as individual receptors or enzymes, to restore immune homeostasis (PubMed, 2022). Therapeutic intervention in this network requires a delicate balance, as broad inhibition can lead to significant safety concerns, including increased susceptibility to infections and impaired wound healing (Journal of Clinical Investigation, 2020).
