Overview

Inflammatory cytokine receptors are a heterogeneous group of transmembrane proteins that serve as the primary sensors for signaling molecules such as IL-4, IL-8, IL-13, IL-17, TNF-alpha, and IFN-alpha. These receptors are categorized into several distinct families, including Type I and II cytokine receptors, the TNF receptor superfamily, and G protein-coupled receptors (GPCRs) for chemokines like IL-8 (UniProt P25024, P25025). Upon ligand binding, they activate complex intracellular pathways, most notably the JAK-STAT, NF-kappaB, and MAPK pathways, which orchestrate immune cell activation, recruitment, and survival (PubMed: 28473537). Dysregulation of these receptors is central to the pathogenesis of chronic inflammatory conditions, including asthma, rheumatoid arthritis, and systemic lupus erythematosus (PubMed: 30272097). Therapeutic targeting of these receptors—using monoclonal antibodies like dupilumab for IL-4Ralpha (UniProt P24394) or anifrolumab for IFNAR1 (UniProt P17181)—has revolutionized the treatment of autoimmune diseases by specifically neutralizing pro-inflammatory signaling. However, because these receptors are essential for host defense, their inhibition carries significant risks, including increased susceptibility to bacterial, viral, and fungal infections (StatPearls: Cytokine Antagonists).
