Target intelligence / Profile preview
Inflammatory cytokines and immune system modulation refers to the broad biological processes and therapeutic strategies centered on signaling proteins that regulate immune responses. Cytokines, such as tumor necrosis factor (TNF), interleukins (IL), and interferons (IFN), act as chemical messengers that coordinate inflammation and host defense (StatPearls: Cytokines, 2023). In many pathological conditions, the dysregulation of these cytokines leads to chronic inflammation and tissue destruction, which are hallmarks of autoimmune diseases like rheumatoid arthritis and psoriasis (Nature Reviews Immunology, 2018). Drugs designed for immune modulation target these pathways by either neutralizing the cytokines directly, blocking their cell-surface receptors, or inhibiting intracellular signaling components like Janus kinases (JAKs). While these therapies have revolutionized the treatment of inflammatory disorders, they carry inherent risks, most notably an increased susceptibility to serious infections and potential malignancies due to the suppression of normal immune surveillance (Journal of Clinical Medicine, 2021). Consequently, this field focuses on achieving a precise balance between dampening harmful inflammation and preserving the patient's ability to respond to pathogens.
Therapeutic modulation involves the neutralization of pro-inflammatory cytokines using monoclonal antibodies, the blockade of cytokine receptors to prevent signal initiation, or the inhibition of intracellular signaling enzymes such as Janus kinases (JAKs) to prevent downstream gene transcription [PMID: 31041429].
