Inflammatory pathway regulators are a broad class of molecular entities that govern the initiation, propagation, and resolution of the inflammatory response (Medzhitov, 2008). This group includes a wide array of targets such as cytokines (e.g., TNF, IL-1, IL-6), their respective receptors, and intracellular signaling molecules like Janus kinases (JAKs) and the NF-kappaB transcription factor complex (Newton & Dixit, 2012). While these pathways are essential for host defense against pathogens, their dysregulation is a primary driver of chronic inflammatory diseases, including rheumatoid arthritis, psoriasis, and inflammatory bowel disease (Furman et al., 2019). Therapeutic strategies targeting these regulators range from monoclonal antibodies that neutralize extracellular ligands to small molecules that inhibit intracellular enzymatic activity (Schett et al., 2013). Because these regulators are central to immune surveillance, their pharmacological modulation often presents significant safety challenges, most notably an increased susceptibility to opportunistic infections and potential long-term risks of malignancy (Singh et al., 2015). Consequently, the term is considered a functional category encompassing multiple distinct therapeutic targets rather than a single specific molecule.
