Overview

An inflammatory state is a complex physiological condition characterized by the activation of the immune system in response to harmful stimuli, such as pathogens, damaged cells, or toxic compounds. It involves the recruitment of leukocytes and the release of various chemical mediators, including cytokines, chemokines, and prostaglandins, which coordinate the body's defense and repair mechanisms (StatPearls, 2023, PMID: 29489241). While acute inflammation is a vital, self-limiting protective process, a chronic inflammatory state occurs when the response is prolonged or dysregulated, leading to progressive tissue destruction and organ dysfunction. This state is a central driver in the pathogenesis of numerous conditions, including rheumatoid arthritis, atherosclerosis, and various cancers (NIH/NCBI, 2023). In clinical pharmacology, the 'inflammatory state' is a phenotypic outcome rather than a specific molecular target; therapeutic interventions aim to resolve this state by targeting specific proteins like TNF-alpha or Janus kinases to mitigate inflammation and prevent long-term damage (Nature Reviews Drug Discovery, 2017).

Mechanism of action

Drugs do not target the 'inflammatory state' as a single molecule; instead, they modulate it by inhibiting specific molecular components such as pro-inflammatory cytokines (TNF-alpha, IL-6), enzymes (COX-1, COX-2), or signaling kinases (JAK1/2/3).
