Overview

IL-13Rα1 is a glycosylated transmembrane protein that serves as a primary subunit of the interleukin-13 receptor complex. It forms functional receptor complexes with IL-4Rα, mediating signaling for both IL-13 and IL-4. It is expressed on a wide range of cell types and plays a central role in Th2-type immune responses, including allergic inflammation, and regulates processes like IgE class switching in B-cells. Dysregulation contributes significantly to diseases characterized by type II inflammation, such as asthma and atopic dermatitis. Therapeutics targeting this axis include monoclonal antibodies like dupilumab that block shared components such as the IL-4Rα subunit.

Mechanism of action

Dupilumab blocks the IL-4Rα subunit, thereby inhibiting signaling from both IL-4 and IL-13 through their common receptors.
