Overview

Interleukin 4 receptor alpha chain is a type I transmembrane receptor subunit encoded by the IL4R gene (CD124). It binds interleukin-4 and interleukin-13, forming receptor complexes crucial for immune regulation. The alpha chain is essential for high-affinity ligand recognition and signal transduction via the JAK/STAT pathway, particularly through STAT6. IL4Rα functions in Th2 cell differentiation, alternative macrophage activation, and regulation of IgE synthesis, with roles in inflammation and allergic responses. Variants and expression levels are associated with atopic diseases, infection susceptibility, and certain cancers. Therapeutically, IL4Rα is targeted by agents like dupilumab for allergy and investigational fusion toxins for oncology.

Mechanism of action

Blockade of IL-4 and IL-13 signaling by antagonism or neutralization of IL4Rα. Inhibition of JAK/STAT-mediated immune responses. Induction of cell death via delivery of cytotoxic agents to IL4Rα-expressing tumor cells.
