Intestinal epithelial cell surface receptors and pattern recognition receptors (PRRs) represent a broad class of sensors that detect pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs) to maintain gut homeostasis (Nature Reviews Gastroenterology & Hepatology, 2013, PMID: 24072361). These include Toll-like receptors (TLRs), NOD-like receptors (NLRs), and C-type lectin receptors (CLRs), which are expressed on or within intestinal epithelial cells and underlying immune cells (Journal of Clinical Investigation, 2017, PMID: 28844788). Upon ligand binding, these receptors assemble into complex signaling scaffolds, such as the Myddosome or inflammasome, to trigger downstream cascades involving Janus kinases (JAKs) and NF-kappaB (Cell, 2018, PMID: 29385115). Dysregulation of these pathways is central to the pathogenesis of inflammatory bowel diseases (IBD), where chronic inflammation leads to mucosal damage (The Lancet, 2017, PMID: 29132860). Therapeutic intervention typically involves targeting specific components of these complexes, such as JAK inhibitors or TLR modulators, to dampen the inflammatory response (Gastroenterology, 2019, PMID: 30545565). However, because these receptors are essential for host defense, their inhibition carries risks of infection and altered tissue repair (Nature Reviews Drug Discovery, 2012, PMID: 22460122).
