Overview

Tofacitinib is a small molecule Janus kinase (JAK) inhibitor that modulates the immune system by blocking the action of JAK enzymes involved in inflammatory signaling pathways. It is primarily used as a disease-modifying anti-rheumatic drug (DMARD) for adults with moderate to severe rheumatoid arthritis, psoriatic arthritis, and ulcerative colitis who have had an inadequate response or intolerance to tumor necrosis factor (TNF) inhibitors. Tofacitinib is also indicated for polyarticular juvenile idiopathic arthritis in children aged 2 years and older and has been approved for ankylosing spondylitis. Developed by Pfizer, it was the first oral JAK inhibitor approved for rheumatoid arthritis[1][2][3][4][5].
