Overview

Janus kinases 1 and 3 (JAK1/JAK3) are intracellular, non-receptor tyrosine kinases that play a central role in cytokine-mediated signal transduction via the JAK-STAT pathway. JAK1 is ubiquitously expressed and associates with various type I/II cytokine receptors, while JAK3 is primarily restricted to hematopoietic cells and specifically binds common gamma chain (γc) containing receptors. Both are essential for proper immune function. Inhibition of JAK1 and JAK3 is a therapeutic strategy for autoimmune diseases, but can lead to increased infection risk due to their central role in immune cell function. Mutations in JAK3 can cause severe combined immunodeficiency (SCID).

Inhibition of JAK1 and/or JAK3 kinase activity, preventing phosphorylation and activation of STAT proteins, thus reducing downstream gene expression related to immune responses and inflammation.
