Overview

Macrophage polarization and inflammatory cytokine signaling describes the dynamic process by which macrophages transition between functional states, primarily the pro-inflammatory M1 and the anti-inflammatory or pro-resolving M2 phenotypes (Nature Reviews Immunology, 2008). This transition is regulated by a complex interplay of intracellular signaling pathways, such as the JAK/STAT and NF-κB pathways, and extracellular signals from cytokines like interferon-gamma, tumor necrosis factor (TNF), and various interleukins (Frontiers in Immunology, 2020). In many chronic diseases, an imbalance in this signaling leads to persistent inflammation or immune evasion, making the components of these pathways high-priority therapeutic targets (Science, 2017). While the term itself refers to a broad biological process rather than a single protein, pharmacological intervention typically involves targeting specific receptors or enzymes within the cascade to restore immune homeostasis (Journal of Clinical Investigation, 2013). Understanding these mechanisms is crucial for developing treatments for conditions ranging from autoimmune disorders to solid tumors, where tumor-associated macrophages often promote progression (Nature, 2019).
