The TNF-α / IL-6 / MMPs and related inflammatory pathways represent a coordinated signaling network central to the innate and adaptive immune responses. Tumor Necrosis Factor-alpha (TNF-α) and Interleukin-6 (IL-6) act as primary pro-inflammatory cytokines that trigger downstream signaling cascades, including the activation of Matrix Metalloproteinases (MMPs) which facilitate tissue remodeling and degradation (Nagase et al., 2006, PMID: 16448344). Dysregulation of this axis is a hallmark of chronic inflammatory conditions such as rheumatoid arthritis, where TNF-α and IL-6 drive joint destruction by inducing MMP expression in synovial fibroblasts (Tanaka et al., 2014, PMID: 25190079). Pharmacological intervention focuses on neutralizing these cytokines or blocking their receptors using monoclonal antibodies like adalimumab or tocilizumab (Idriss & Naismith, 2000, PMID: 11071201). While these therapies are highly effective in reducing systemic inflammation, they are associated with significant safety concerns, most notably an increased susceptibility to serious infections and potential long-term risks of malignancy (StatPearls, NBK470413).
