The pro-inflammatory and anti-inflammatory cytokine network is a complex regulatory system of signaling proteins that coordinate the body's immune and inflammatory responses. Pro-inflammatory cytokines, such as Tumor necrosis factor alpha, Interleukin 1, and Interleukin 6, are secreted by immune cells to initiate defense mechanisms against pathogens and injury (StatPearls, 2023). To prevent uncontrolled inflammation, anti-inflammatory cytokines like Interleukin 10 and Transforming growth factor beta are produced to dampen the response and facilitate tissue repair (Nature Reviews Immunology, 2018). A persistent imbalance in this network, often characterized by an overproduction of pro-inflammatory mediators, is a primary driver of chronic autoimmune diseases like rheumatoid arthritis and Crohn's disease, as well as acute life-threatening conditions like cytokine release syndrome (PubMed, 2021). Therapeutic interventions frequently target specific components of this network using monoclonal antibodies, decoy receptors, or small molecule inhibitors to restore homeostatic balance (NIH, 2022). Because this term refers to a broad physiological framework involving dozens of distinct proteins rather than a single molecular entity, it is classified as a biological network rather than a specific therapeutic target.
Modulation of the cytokine balance through neutralization of pro-inflammatory ligands, blockade of cytokine receptors, or inhibition of downstream signaling pathways like the Janus kinase/Signal transducer and activator of transcription (JAK/STAT) pathway.
