Overview

Receptor Tyrosine-Protein Kinase erbB2 (HER2) is a member of the human epidermal growth factor receptor (HER/EGFR/ERBB) family and a receptor tyrosine kinase. Unlike other EGFR family members, HER2 does not directly bind ligands but is activated by homo- or heterodimerization with other ERBB receptors, leading to autophosphorylation and activation of downstream signaling pathways such as MAPK and PI3K. HER2 plays key roles in cell growth, differentiation, and survival. Overexpression or amplification of HER2 is observed in a significant percentage of breast and gastric cancers, driving aggressive tumor behavior and serving as a crucial biomarker for targeted therapies like trastuzumab and lapatinib.
