Overview

The Signal Transducer and Activator of Transcription (STAT) signaling pathway is a fundamental intracellular mechanism that converts extracellular signals from cytokines and growth factors into specific gene expression programs (Source: NIH, PubMed). The pathway is typically initiated by the binding of ligands to cell-surface receptors, which activates associated Janus kinases (JAKs) to phosphorylate the receptors and subsequently the STAT proteins themselves (Source: StatPearls). Once phosphorylated, STAT proteins form homo- or heterodimers and translocate into the nucleus, where they bind to promoter regions of target genes to regulate cell growth, survival, and immune function (Source: UniProt). Dysregulation of this pathway, particularly the constitutive activation of STAT3 and STAT5, is a hallmark of many human cancers and inflammatory diseases, contributing to tumor progression, metastasis, and immune evasion (Source: Nature Reviews Cancer).
