The Signal Transducer and Activator of Transcription (STAT) family consists of seven intracellular transcription factors (STAT1, STAT2, STAT3, STAT4, STAT5A, STAT5B, and STAT6) that mediate the cellular responses to cytokines, hormones, and growth factors (Source: UniProt, P40763; StatPearls, NBK545175). Upon activation by Janus kinases (JAKs) through phosphorylation, STAT proteins dimerize and translocate to the nucleus to regulate the expression of genes involved in critical processes such as cell growth, survival, and immune function (Source: PubMed, 30072362). Dysregulation of the STAT pathway, particularly constitutive activation of STAT3 and STAT5, is a hallmark of many cancers and inflammatory diseases, making these proteins high-priority therapeutic targets (Source: PubMed, 31039559). While many clinical strategies currently focus on upstream JAK inhibitors, direct STAT inhibitors—including small molecules targeting the SH2 domain and antisense oligonucleotides—are actively being developed to achieve greater specificity and reduced systemic toxicity (Source: PubMed, 32883704). Therapeutic challenges include the high structural homology between family members and the necessity of maintaining basal signaling for normal physiological functions (Source: PubMed, 29439339).
