Overview

The Th2 cytokine production pathway describes the cascade of molecular events and cellular differentiation that lead to the generation of Th2 cells, a subset of CD4+ T helper cells. Upon stimulation by cytokines, particularly interleukin-4 (IL-4), naïve CD4+ T cells differentiate into Th2 cells with the aid of antigen-presenting cells and transcription factors such as GATA3 and STAT6[2][3][6]. Mature Th2 cells secrete cytokines including IL-4, IL-5, and IL-13, which are essential in orchestrating humoral immunity, driving IgE class-switching in B cells, recruiting and activating eosinophils, and defending against helminth infections[2][5]. Dysregulation of this pathway contributes to the pathogenesis of allergic and type 2 inflammatory diseases, including asthma and atopic dermatitis. This pathway is targeted indirectly by drugs that block the activity of Th2 cytokines or their signaling receptors to manage such diseases[5][6].
