Overview

The T helper type 2 cell signaling pathway encompasses the activation, differentiation, and function of Th2 cells, a subset of CD4+ T cells distinguished by their production of cytokines such as IL-4, IL-5, IL-9, and IL-13, which drive humoral immunity and tissue repair. Th2 differentiation is governed by stimuli such as IL-4/IL-2 signaling, GATA3 transcriptional activation, and STAT6/STAT5-mediated signal transduction. Th2 cells orchestrate protective responses against extracellular parasites and allergens, modulate tissue repair, but are implicated in the pathogenesis of allergic diseases (e.g., asthma, atopic dermatitis) and fibrotic disorders when dysregulated. Therapeutic strategies often focus on blocking Th2 cytokines or downstream signaling to treat related diseases, but careful regulation is necessary to avoid impeding beneficial tissue repair or inducing adverse immune effects.
