Overview

The **JAK-STAT signaling pathway** is a critical mediator of cellular responses to cytokines and growth factors. **Janus kinase 2 (JAK2)** is a cytoplasmic non-receptor tyrosine kinase that phosphorylates and activates **signal transducer and activator of transcription 3 (STAT3)**, a transcription factor. Upon extracellular stimulation (such as by interleukin-6), JAK2 is activated and subsequently phosphorylates STAT3, which then dimerizes, translocates to the nucleus, and regulates gene expression essential for cell proliferation, survival, differentiation, and immune response[1][4][5][6][7]. Dysregulation of the JAK2/STAT3 signaling axis contributes to various diseases, particularly cancers and inflammatory disorders, leading to the development of numerous drugs targeting either JAK2 or STAT3 or their upstream activators[2][5][6].

Mechanism of action

Inhibition of kinase activity to prevent STAT3 phosphorylation (JAK2 inhibitors); Blockade of STAT3 dimerization or DNA binding; Inhibition of upstream cytokines or receptors to suppress pathway activation (e.g., IL-6 pathway blockade); Induction of degradation of STAT3 protein
