This complex is a central driver of the inflammatory cascade, and its overactivity is implicated in the pathogenesis of various autoimmune diseases, including rheumatoid arthritis, Crohn's disease, and psoriasis (StatPearls NBK482425). Pharmacological intervention typically involves the use of biologics, such as monoclonal antibodies or decoy receptors, which prevent the formation of the complex by sequestering TNF-α (NIH/NCBI Bookshelf).
