Overview

Tumour necrosis factor (TNF), formerly known as TNF-α, is a pro-inflammatory cytokine and a key chemical messenger produced by the immune system. It plays a central role in mediating inflammation and orchestrates various physiological and pathological processes, including immune cell activation, fever induction, apoptosis (programmed cell death), necroptosis (regulated necrosis), and tissue homeostasis. Excessive or dysregulated production of TNF is implicated in several diseases, including chronic inflammatory diseases like rheumatoid arthritis, psoriasis, and inflammatory bowel disease, as well as cancer and neurodegenerative disorders. Therapeutics targeting TNF are widely used to treat autoimmune/inflammatory disorders by blocking its interaction with receptors.
