Overview

ZNF649 antisense RNA 1 (ZNF649-AS1) is a long non-coding RNA found on chromosome 19 that acts as an antisense transcript relative to the ZNF649 gene. It is upregulated in trastuzumab-resistant breast cancer cells and tissues, and its high expression is associated with poor response and shorter survival in patients receiving trastuzumab therapy. Functionally, ZNF649-AS1 promotes autophagy and suppresses apoptosis by recruiting the RNA binding protein PTBP1, which enhances the transcription of ATG5, an autophagy-related gene. Inhibition of ZNF649-AS1 can reverse trastuzumab resistance and sensitize cancer cells to therapy, making it a potential therapeutic target and biomarker for monitoring treatment efficacy in HER2-positive breast cancer patients. Its precise biological roles are still being elucidated, but it is implicated in the post-transcriptional regulation of cell survival and drug response pathways.
