Overview

ABL103 is a novel T-cell engaging bispecific antibody developed by ABL Bio. It is designed to simultaneously target B7-H4, an immune checkpoint molecule expressed on tumor cells, and 4-1BB (CD137), a costimulatory receptor on T cells. The drug’s mechanism of action involves B7-H4-dependent activation of 4-1BB signaling pathways within the tumor microenvironment, leading to selective activation and proliferation of T cells only in the presence of B7-H4-expressing tumor cells. This approach aims to enhance anti-tumor immunity while minimizing off-tumor toxicity commonly associated with systemic 4-1BB agonists. Preclinical studies have shown that ABL103 induces potent anti-tumor activity and immunological memory, completely eliminating tumors in advanced models and protecting against rechallenge. The drug is being developed for advanced or metastatic solid tumors, including triple-negative breast cancer and other B7-H4-positive cancers[1][2][3][5][6][8].
