Abrocitinib is an oral small molecule drug developed by Pfizer that acts as a selective Janus kinase 1 (JAK1) inhibitor. It works by reversibly and selectively blocking the ATP-binding site of JAK1, thereby inhibiting the signaling of multiple cytokines involved in immune and inflammatory responses. This mechanism modulates pathways associated with atopic dermatitis pathophysiology, including interleukins IL-4, IL-13, IL-31, IL-22 and thymic stromal lymphopoietin (TSLP). Abrocitinib is primarily indicated for the treatment of moderate to severe atopic dermatitis in adults and adolescents aged 12 years or older who have not responded adequately to other systemic therapies or for whom such therapies are not advisable[1][2][3][4][5].
