ADS051

ADS051 is a novel, oral, gut-restricted small molecule modulator of neutrophil trafficking and activation. It is based on a modified cyclosporine scaffold covalently linked to a 2,000 molecular weight polyethylene glycol (PEG) by an amide linker. ADS051 works by inhibiting multidrug resistance protein 2 (MRP2) and formyl peptide receptor 1 (FPR1), which modulates neutrophil trafficking and activity. Unlike traditional immunosuppressants, it is designed to be gut-restricted with minimal systemic exposure, targeting neutrophil-mediated tissue damage in the colon while limiting general immunosuppression. It is being developed primarily for the treatment of ulcerative colitis.
