AHA-1031 is an Fc-engineered, ADCC-enhanced monospecific monoclonal antibody developed by Aakha Biologics for the treatment of advanced non-small cell lung cancer (NSCLC). The antibody targets Major Histocompatibility Complex class I-related chain A and B (MICA/B), which are stress-induced ligands expressed on the surface of various tumor cells. In the tumor microenvironment, these ligands are often proteolytically shed to evade immune detection; AHA-1031 works by specifically blocking this shedding process while maintaining the ligands' ability to bind the NKG2D receptor on natural killer (NK) cells and cytotoxic T cells. The engineered Fc region further enhances antibody-dependent cellular cytotoxicity (ADCC), facilitating the recruitment and activation of immune cells to eliminate the cancer. Currently in IND-enabling studies, AHA-1031 has shown significant monotherapy activity in preclinical models of KRAS-LKB1 mutant lung cancer.
