anifrolumab + voclosporin

Anifrolumab + voclosporin is a combination therapy consisting of two distinct immunomodulatory agents used in the management of systemic lupus erythematosus (SLE), particularly in patients with both lupus nephritis (LN) and cutaneous manifestations such as discoid lupus erythematosus (DLE). Anifrolumab is a fully human IgG1κ monoclonal antibody that targets and inhibits the type I interferon receptor subunit 1 (IFNAR1), thereby blocking type I interferon signaling, which plays a central role in SLE pathogenesis. Voclosporin is a calcineurin inhibitor that suppresses T-cell activation by inhibiting calcineurin, leading to reduced transcription of early inflammatory cytokines and stabilization of podocytes in the kidney. The combination has been reported to allow for steroid-sparing regimens and may provide additive or complementary benefits for patients with refractory disease manifestations[1][2][10].
