α-CD137 antibody (also known as anti-CD137 or anti-4-1BB) is an agonistic monoclonal antibody designed to target and activate the CD137 receptor, a member of the tumor necrosis factor receptor (TNFR) superfamily. CD137 is expressed on the surface of activated T cells, natural killer (NK) cells, and certain myeloid cells, where it functions as a potent co-stimulatory molecule. Binding of the antibody to CD137 triggers signaling pathways that promote T-cell proliferation, survival, and effector functions, such as cytokine production and cytotoxicity. In the context of cancer immunotherapy, α-CD137 antibodies are utilized to stimulate a robust and durable antitumor immune response, often demonstrating synergistic effects when combined with other modalities like intratumoral vaccines (e.g., CisVac) or checkpoint inhibitors. Preclinical studies have shown that this approach can lead to the activation of tumor antigen-specific CD8+ T cells and the remodeling of the tumor microenvironment to overcome immune suppression.
