An anti-CXCL1 neutralizing antibody is a monoclonal antibody designed to bind specifically to Chemokine (C-X-C motif) ligand 1 (CXCL1), also known as GRO-alpha, and inhibit its biological activity. CXCL1 is a pro-inflammatory chemokine that signals primarily through the CXCR2 receptor to mediate the chemoattraction and activation of neutrophils. It plays a significant role in various pathological processes, including chronic inflammation, angiogenesis, and tumor progression, where it promotes tumor cell proliferation, metastasis, and the recruitment of myeloid-derived suppressor cells (MDSCs). By neutralizing CXCL1, these antibodies aim to disrupt the CXCL1/CXCR2 signaling axis, thereby reducing pathological inflammation and inhibiting tumor growth. While widely utilized as research tools (such as clone 124014) to study chemokine biology in models of cancer and inflammatory disease, clinical development of specific anti-CXCL1 therapeutic assets has been limited compared to small molecule inhibitors of the CXCR2 receptor.
