Anti-HMW-MAA IgG4 antibody is a recombinant monoclonal antibody of the IgG4 subclass directed against the High Molecular Weight Melanoma Associated Antigen (HMW-MAA), also known as Chondroitin Sulfate Proteoglycan 4 (CSPG4). Developed by researchers at King's College London and the Institute of Cancer Research, this antibody was engineered as part of a comparative study to evaluate the functional properties of different antibody classes (IgG1, IgG4, and IgE) in melanoma immunotherapy. HMW-MAA is a membrane-bound protein overexpressed in more than 90% of melanomas and is implicated in tumor cell proliferation, migration, and metastasis. The IgG4 variant was designed to engage specific Fc receptors on immune effector cells, such as monocytes, to induce antibody-dependent cell-mediated phagocytosis (ADCP) and cytotoxicity (ADCC). Preclinical studies in NOD/SCID mice indicated that while the IgG4 antibody showed activity, its IgE counterpart demonstrated superior anti-tumor efficacy.
