Anti-IL-4Ralpha antibodies are a class of therapeutic monoclonal antibodies designed to bind to the alpha subunit of the interleukin-4 receptor (IL-4Rα). This receptor subunit is a critical component of both the Type I and Type II IL-4 receptor complexes, which mediate the signaling of interleukin-4 (IL-4) and interleukin-13 (IL-13). These cytokines are central drivers of Type 2 (Th2) helper T-cell-mediated inflammation, which underlies the pathogenesis of various allergic and atopic conditions. By blocking IL-4Rα, these antibodies effectively inhibit the dual signaling pathways of IL-4 and IL-13, leading to a reduction in inflammatory markers and clinical symptoms. The most prominent member of this class is dupilumab, which has received regulatory approval for multiple indications including atopic dermatitis, asthma, and chronic rhinosinusitis. In preclinical research, such as mouse models of atopic dermatitis, anti-IL-4Ralpha antibodies (often murine surrogates) are frequently used as benchmark treatments to evaluate the efficacy of novel anti-inflammatory agents.
