Selonterra's APOE4 modulator is an investigational small molecule therapeutic program designed to treat Alzheimer's disease by addressing the genetic risk associated with the APOE4 allele. Unlike conventional approaches that target the APOE4 protein itself, Selonterra's strategy is based on proprietary findings that the APOE4 DNA sequence acts as a regulatory element, controlling the expression of a network of genes in its chromosomal vicinity. By modulating this gene expression, the small molecule aims to correct the underlying biological dysfunction in human neurons caused by the APOE4 variant. The program is currently in the IND-enabling stage, representing a novel mechanism of action distinct from amyloid or tau-targeting therapies.
