Overview

ARQ-252 is a potent and highly selective topical small molecule inhibitor of Janus kinase type 1 (JAK1), developed for the treatment of inflammatory dermatologic diseases such as chronic hand eczema and vitiligo. Its mechanism centers on blocking JAK1, a key enzyme in the signaling pathways of many inflammatory cytokines involved in immune system function. The drug is distinguished by its high selectivity for JAK1 over JAK2, which may reduce hematopoietic side effects commonly associated with less selective JAK inhibitors. The active pharmaceutical ingredient in ARQ-252 is SHR0302, which has also been studied as an oral agent (ivarmacitinib) for systemic inflammatory conditions including rheumatoid arthritis and atopic dermatitis[1][2][5][6][9].
