Autologous oncolysate-pulsed dendritic cell vaccine is an individualized cancer immunotherapy consisting of a patient's own dendritic cells (DCs) loaded with lysates derived from their own tumor cells (oncolysates). The DCs are typically differentiated in vitro from peripheral blood monocytes and then 'pulsed' with the tumor lysate, which contains a broad spectrum of patient-specific tumor-associated antigens. Once administered back into the patient, these mature, antigen-presenting DCs migrate to lymphoid organs where they prime and activate tumor-specific CD4+ helper and CD8+ cytotoxic T-lymphocytes. This process aims to induce a robust, multi-antigenic immune response capable of identifying and eliminating residual or metastatic cancer cells. Clinical studies, including phase I/II trials in colorectal cancer, have demonstrated the vaccine's safety and potential for long-term relapse-free survival.
